SDCAA Medium is a defined, phosphate-buffered medium formulated for cultivation and display workflows in yeast. It contains glucose as the primary carbon source, yeast nitrogen base without amino acids and with ammonium sulfate as the inorganic nitrogen and micronutrient source, casamino acids as a complex amino acid supply, and a disodium/monosodium phosphate system for buffering.

This formulation is suitable for growth and maintenance of auxotrophic Saccharomyces cerevisiae strains when used with appropriate dropout supplements and selection systems. For laboratory use in microbiology.

  • 2.0% Cerelose Anhydrous Dextrose (Glucose)
  • 0.67% Yeast Nitrogen Base (w/o AA and w/AS)
  • 0.50% Casamino Acids
  • 0.54% Disodium Phosphate
  • 0.86% Monosodium Phosphate
  • Yeast Cultivation and Propagation: Defined medium for robust growth of Saccharomyces cerevisiae and related yeasts with controlled carbon and nitrogen sources.

  • Yeast Surface Display Workflows: Standard growth medium for library expansion and selection prior to galactose-based induction.

  • Auxotrophic Selection Assays: Supports selection schemes leveraging casamino acids (low in tryptophan) with YNB (w/o amino acids, w/ ammonium sulfate).

  • Glucose-Repression and Metabolic Studies: 2% glucose enables controlled repression of GAL promoters for regulatory and metabolic experiments.

  • Protein Expression Screening in Yeast: Used for pre-induction growth and plasmid maintenance in cloning and expression workflows.

  • Quantitative Growth and Fitness Assays: Chemically defined formulation enables reproducible OD-based growth curves and competition assays.

  • Phosphate-Buffered, pH-Stable Culturing: Disodium and monosodium phosphate provide buffering for consistent pH during cultivation and assays.

  • Strain Banking and Starter Cultures: Suitable for preparing glycerol stocks and inocula for downstream yeast experiments.
