What is LB broth?

LB broth, also known as Luria-Bertani Broth or Lysogeny Broth, is a nutrient-rich medium widely used in microbiology for the rapid growth and cultivation of many bacterial species, particularly E. coli. 

 

LB liquid media contains three primary components: tryptone, yeast extract, and sodium chloride. Tryptone supplies essential amino acids and peptides that serve as accessible nitrogen and carbon sources for bacterial protein synthesis and growth. Yeast extract provides a complex mixture of water-soluble vitamins, trace elements, and organic compounds, including B vitamins and nucleotides, which support the metabolic functions of the bacteria. Sodium chloride maintains osmotic balance and supplies sodium ions, which are essential for cellular transport mechanisms, ensuring a stable environment for bacterial proliferation.

Lysogeny Broth (LB) is a nutritionally rich bacterial growth medium used for cultivating E. coli and other non-fastidious microorganisms. It’s commonly used in molecular biology workflows such as plasmid propagation, protein expression, and cloning.

Standard LB formulations contain 10 g of tryptone, 5 g of yeast extract, and 10 g of sodium chloride per liter. Variants such as LB Lennox Broth contain 5 g/L of sodium chloride, which can affect bacterial growth and recombinant protein yield.

Antibiotics are commonly added to LB media to maintain selective pressure for plasmid-containing or genetically modified bacterial strains. We carry antibiotic-free LB broth and formulations supplemented with ampicillin (50 µg/mL and 100 µg/mL), carbenicillin (50 µg/mL and 100 µg/mL), kanamycin (25 µg/mL and 50 µg/mL), Zeocin (25 µg/mL), and chloramphenicol (25 µg/mL). LB broth is a liquid medium that supports the growth of bacteria in suspension for applications like cloning or protein expression, while LB agar contains added agar to create a solid surface for isolating and maintaining individual bacterial colonies. Both share the same nutrient base, including tryptone, yeast extract, and sodium chloride, but differ in their physical state and use case. We sell liquid LB broth in tubes, bottles, and bags, as well as bottled LB agar (1.5% agar) and LB top agar (0.8% agar).
