What is 2xYT broth?

2xYT broth is a non-selective, nutrient-dense bacterial growth medium composed of yeast extract, tryptone, and sodium chloride. This enriched composition provides elevated levels of amino acids, peptides, vitamins, and growth factors, supporting faster growth rates and higher cell densities than LB liquid media.


2xYT broth is most commonly used to cultivate E. coli and other non-fastidious bacteria for applications such as high-efficiency cloning, plasmid amplification, recombinant protein production, phage propagation, and phage display. 2xYT broth can be used as a non-selective liquid medium or supplemented with antibiotics to support the selective growth of genetically engineered bacterial strains and for molecular cloning. Another formulation, 4xYT, has twice the concentrations of sodium chloride, yeast extract, and tryptone as 2xYT and is used for rapid bacterial growth and higher cell densities. Glucose can also be added to 2xYT to prevent the leaky expression of toxic recombinant proteins controlled by the lac promoter through catabolite repression.
