Cell Lysis and Endonuclease Treatment

Efficient cell lysis and endonuclease treatment are essential for maintaining high AAV yields and purity. We have proven cell lysis buffers in stock in multiple sizes to match your bioreactor needs, as well as Benzonase® Nuclease to reduce product related impurities such as residual host-cell DNA and plasmid.

 

Our Mammalian Cell Lysis Buffer is a polysorbate 20-based cell lysis reagent at a 10X concentration. The 100 mL volume is sufficient for 1 L of cell culture harvest, and the 500 mL volume is sufficient for 5 L of cell culture harvest.

Ultracentrifugation

To purify your AAV, add the clarified cell lysate to a discontinuous iodixanol gradient. The 60% and 25% gradients contain phenol red to facilitate visualization of the different phases. Lower density contaminants, including empty AAV capsids, will gravitate toward the upper gradients (15-40%) during ultracentrifugation, while the 60% iodixanol gradient serves as a cushion for the full genome containing AAV capsids.

 

Tip: Keeping iodixanol gradient solutions refrigerated can make it easier to layer the solutions.

 

Our off-the-shelf iodixanol gradients include 15% iodixanol, 25% iodixanol with or without phenol red, 40% iodixanol, and 60% iodixanol with phenol red. 0.5% phenol red, PBS-MK, and PBS-MK with 1 M NaCl are also available to tailor your own gradient solutions.

Filtration buffer
Diafiltration and Concentration

For easy AAV diafiltration and concentration when using either a centrifugal filter or tangential flow filtration, we offer PBS and Poloxamer 188 concentrates so you can easily tailor your own solution.

 

TIP: multiple buffer exchanges may be required to completely remove the iodixanol.
