What is 2xYT agar?

2xYT agar plates are a nutrient-rich, general-purpose, solid bacterial culture medium composed of yeast extract, tryptone, sodium chloride, and agar. This enriched composition provides elevated levels of amino acids, peptides, vitamins, and growth factors compared to LB agar, thereby supporting increased bacterial biomass and faster growth.


2xYT agar plates are commonly used for cultivating E. coli and other non-fastidious bacteria in applications such as cloning, plasmid amplification, strain maintenance, recombinant protein expression, phage propagation, and phage display. 2xYT agar may be used as a non-selective growth medium or supplemented with antibiotics for selection during genetic manipulation and molecular cloning experiments.

2xYT is a yeast extract-tryptone agar medium used to culture non-fastidious bacteria, particularly E. coli. It is commonly used for cloning, plasmid propagation, recombinant protein expression, phage propagation, phage display, and other molecular biology applications that benefit from increased nutrient availability.

2xYT agar plates primarily support the growth of non-fastidious bacteria, including laboratory strains of E. coli commonly used in genetic engineering, recombinant protein expression, and molecular cloning workflows.

Both are yeast extract-tryptone-based media, but 2xYT agar contains higher concentrations of yeast extract and tryptone than LB agar, which can support faster growth rates, higher cell densities, and, in some cases, increased plasmid yield.

We sell 2xYT agar plates in non-selective and selective formulations. Our selective 2xYT agar media includes carbenicillin (50µg/mL and 100µg/mL), kanamycin (30µg/mL and 50µg/mL), ampicillin (100µg/mL), tetracycline (20µg/mL), chloramphenicol (34µg/mL), and various combinations.
