LB Broth (Lysogeny Broth) is a nutrient medium used for cultivation and maintenance of laboratory strains of Escherichia coli and other enteric bacteria. It supplies peptides, amino acids, vitamins, and sodium ions to support cellular metabolism and maintain osmotic balance in liquid culture.

Applications include routine propagation, starter cultures for downstream workflows, and selection of plasmid-bearing transformants when supplemented with appropriate antibiotics.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • Routine Bacterial Cultivation: General-purpose liquid medium for growing non-fastidious bacteria such as Escherichia coli and related enteric species.

  • Plasmid Propagation and Cloning: Supports high-density growth for plasmid maintenance and amplification in E. coli-based cloning workflows.

  • Antibiotic Selection in Liquid Culture: Used with selective antibiotics to maintain plasmids or selectable markers in E. coli cultures.

  • Starter and Overnight Cultures: Preparation of inocula for downstream applications including cloning, sequencing, and protein expression.

  • Transformation Recovery/Outgrowth: Short post-transformation recovery of E. coli prior to plating or selection.

  • Protein Expression Pre-cultures: Growth of seed cultures of recombinant E. coli for subsequent induction in production media.

  • Glycerol Stock Preparation: Culturing cells prior to cryopreservation of E. coli strains and plasmid libraries.

  • Educational and QC Applications: Standard medium for teaching labs and routine strain maintenance/verification in microbiology.
