Mueller Hinton II Agar Plates are cation-adjusted agar media formulated for standardized antimicrobial susceptibility testing by disk diffusion and dilution methods. The medium contains an acid hydrolysate of casein, beef extract, and soluble starch, with agar as the gelling agent, and defined levels of calcium and magnesium.

Used for testing nonfastidious aerobic bacteria commonly evaluated in AST, including Escherichia coli, Staphylococcus aureus, Enterococcus faecalis, and Pseudomonas aeruginosa.

  • 1.750% Casamino Acids
  • 0.300% Beef Extract
  • 0.150% Soluble Starch
  • 1.700% Agar
  • 0.002% Calcium Chloride Dihydrate
  • 0.001% Magnesium Chloride Hexahydrate
  • Antimicrobial Susceptibility Testing (Kirby–Bauer Disk Diffusion): Standardized, cation-adjusted medium for reproducible inhibition zones with organisms like Escherichia coli, Staphylococcus aureus, Pseudomonas aeruginosa, and Enterococcus faecalis.

  • Gradient Diffusion MIC (Etest): Determines minimum inhibitory concentrations on uniform lawns of nonfastidious bacteria.

  • Antibiotic Potency and QC Testing: Used with reference strains (e.g., Escherichia coli ATCC 25922, Staphylococcus aureus ATCC 25923, Pseudomonas aeruginosa ATCC 27853, Enterococcus faecalis ATCC 29212).

  • Cultivation and Isolation of Nonfastidious Bacteria: Supports robust growth for routine culture and subculture of organisms such as Escherichia coli and Staphylococcus aureus.

  • Colony Enumeration and Purity Checks: Used for CFU counts, streak isolation, and verification of culture purity from clinical, environmental, or food samples.

  • Antimicrobial Peptide/Bacteriocin Assays (Spot-on-Lawn): Evaluates activity of antibiotics and peptides against standardized bacterial lawns.

  • Combination/Synergy Testing: Assesses interactions of antimicrobial agents using disk proximity or strip overlays on consistent growth medium.

  • Testing Fastidious Organisms with Supplements: Basis for HTM-like applications when supplemented (e.g., susceptibility testing of Haemophilus influenzae and Neisseria gonorrhoeae).
