Minimal M9CA Broth is a defined minimal medium containing a carbohydrate source, an amino acid supplement, a vitamin cofactor, and divalent cations in an M9 salts base. It is used for the cultivation of laboratory strains of E. coli and other enteric bacteria under controlled nutrient conditions.

This formulation supports growth of prototrophic and auxotrophic strains and is suitable for applications requiring defined media. It can be used for maintenance of plasmid-bearing cultures when appropriate selective agents are added by the user.

  • 1X M9 Salts
  • 1.0% Cerelose Anhydrous Dextrose (Glucose)
  • 0.1% Casamino Acids
  • 0.5µg/mL Thiamine
  • 0.2mM Magnesium Sulfate
  • 0.1mM Calcium Chloride
  • Defined Growth of Escherichia coli: Routine cultivation in a minimal, glucose-defined medium supplemented with casamino acids and thiamine.

  • Auxotroph Selection and Screening: Identification of amino acid auxotrophies (commonly leveraging casamino acids that are low in tryptophan) in enteric bacteria.

  • Recombinant Protein Production: Controlled expression in E. coli under minimal conditions to reduce background metabolites and improve reproducibility.

  • Metabolomics and Flux Analysis: Experiments requiring defined nutrients (1X M9 salts, glucose, Mg2+, Ca2+, thiamine) for consistent metabolic baselines.

  • Plasmid Maintenance with Antibiotic Selection: Stable growth of recombinant strains in a chemically defined medium for consistent selection pressure.

  • Carbon Source Utilization Studies: Assessment of glucose-dependent growth phenotypes and catabolite repression effects in E. coli.

  • Cultivation of Other Enteric Bacteria: Growth of organisms such as Salmonella and Shigella in a defined broth with essential ions (Mg2+, Ca2+).
