SDCAA Agar Plates are a synthetic, phosphate-buffered dextrose–casamino acid medium formulated for cultivation and plating of yeast. Glucose serves as the primary carbon source, yeast nitrogen base supplies essential inorganic salts and vitamins, and casamino acids provide a defined mixture of amino acids. Agar provides a solid surface for colony isolation. Sorbitol is included as an osmotic stabilizer, and the disodium/monosodium phosphate pair maintains stable pH during growth.

Suitable for routine propagation, colony isolation, and maintenance of laboratory yeast strains, including Saccharomyces cerevisiae, in applications requiring defined nutrient composition and buffering.

  • 1M Sorbitol
  • 2.00% Glucose
  • 0.67% Yeast Nitrogen Base
  • 0.50% Casamino Acids
  • 0.54% Disodium Phosphate
  • 0.86% Monosodium Phosphate
  • 1.50% Agar
  • Yeast Transformation Recovery and Plating: Osmotically stabilized outgrowth and plating of Saccharomyces cerevisiae after electroporation or spheroplasting (1 M sorbitol; phosphate-buffered).

  • Yeast Surface Display Library Handling: Library outgrowth, titering, and plating prior to induction in SGCAA.

  • Clonal Isolation and Colony Picking: Defined, glucose-based medium for isolating single yeast colonies.

  • CFU Determination and Plating Efficiency: Quantification of yeast viability and transformation efficiency on 2.00% glucose plates.

  • Propagation of Auxotrophic Strains: Growth of auxotrophic yeast carrying plasmids in a casamino acid–supplemented, phosphate-buffered environment.

  • Phenotypic and Binding Screens on Plates: Assays requiring consistent composition (0.67% YNB, 0.50% casamino acids) and stable pH (Na2HPO4/NaH2PO4).

  • Spheroplast Support: Recovery and maintenance of fragile yeast cells using 1 M sorbitol for osmotic stabilization.

  • Defined Medium Applications: Experiments avoiding complex nutrients, leveraging 2.00% glucose and 1.50% agar for reproducible growth.
