SDCAA Agar Plates are a defined agar medium used for cultivation, plating, and maintenance of Saccharomyces cerevisiae strains, including applications such as yeast display workflows, clone propagation, and library screening under auxotrophic selection.

The medium provides a glucose carbon source, an osmotic stabilizer, a phosphate buffering system, yeast nitrogen base with ammonium for inorganic nutrients and vitamins, and casamino acids for readily accessible amino acids, supporting consistent growth and colony formation of defined yeast strains.

  • 1M Sorbitol
  • 0.540% Disodium Phosphate
  • 0.856% Monosodium Phosphate Monohydrate
  • 1.500% Agar
  • 2.000% Glucose
  • 0.671% Yeast Nitrogen Base with Ammonium Sulfate
  • 0.500% Casamino Acids
  • Yeast Cultivation and Plating: Supports robust growth of Saccharomycetales using glucose, YNB with ammonium sulfate, and casamino acids.

  • Transformant Recovery: 1 M sorbitol provides osmotic stabilization for recovery after lithium acetate/PEG or electroporation-based yeast transformations.

  • Spheroplast Handling: Maintains viability of yeast spheroplasts during plating and recovery.

  • Colony Isolation and Titering: Enables single-colony isolation and CFU determination for yeast libraries and clones.

  • Yeast Display Workflows: Used for growth under glucose-mediated GAL promoter repression prior to induction on galactose media.

  • Selective Screening with TRP1: Casamino acids (tryptophan-poor) permit TRP1-based selection while complementing most other auxotrophies.

  • Cell Wall Mutant Phenotyping: Osmotic support facilitates screening of cell wall–defective or osmo-fragile yeast strains.

  • Replica Plating and Strain Purification: Suitable for transferring and purifying yeast strains across plates.

  • Pichia pastoris Workflows: Supports transformation recovery and clone screening in methylotrophic yeast.
