SDCAA Agar Plates with Tryptophan-40 are a synthetic defined, casamino acid–based agar medium for cultivation and plating of Saccharomyces cerevisiae, including auxotrophic strains and spheroplast recovery workflows.

Glucose at 2.00% serves as the primary carbon source; yeast nitrogen base and casamino acids provide defined nutrients; a phosphate buffer system stabilizes pH; 1 M sorbitol supplies osmotic support; 1.50% agar solidifies the medium; and 40 µg/mL tryptophan provides defined supplementation for trp auxotrophy and selection schemes.

  • Yeast Transformation Recovery: Osmotically stabilized (1 M sorbitol) plates for recovering spheroplasts and transformants.

  • Auxotrophic Selection (Non-TRP): Defined SD-CAA with 40 µg/mL tryptophan for selecting/maintaining strains using markers like LEU2, HIS3, URA3, or LYS2.

  • Colony Isolation and Streaking: Plating, single-colony isolation, and colony counting of Saccharomyces cerevisiae on 2.00% glucose with 0.67% YNB and 0.50% CAA.

  • Yeast Surface Display: Library plating and clonal picking on defined SD-CAA for display workflows.

  • CRISPR/Cas9 Editing Workflows: Post-editing selection and recovery on defined medium supporting auxotrophic selection.

  • Phenotype and Drug Sensitivity Assays: Growth assays on a defined background (phosphate-buffered, 1.50% agar) for comparative studies.
