SOC Medium is a nutrient-rich broth that supports recovery and outgrowth of transformed E. coli cells. Its composition provides conditions suitable for metabolic recovery and plasmid expression prior to selective plating.

  • 2.0% Tryptone
  • 0.5% Yeast Extract
  • 10mM Sodium Chloride
  • 2.5mM Potassium Chloride
  • 10mM Magnesium Chloride
  • 10mM Magnesium Sulfate
  • 20mM Cerelose Anhydrous Dextrose (Glucose)
  • Recovery after Chemical Transformation: Short outgrowth of transformed E. coli post heat-shock to allow membrane repair and expression of antibiotic resistance.

  • Outgrowth after Electroporation: Immediate recovery medium for electroporated E. coli, supporting membrane repair and high viability.

  • Maximizing Transformation Efficiency: Enhances plasmid uptake outcomes in competent E. coli workflows for cloning.

  • Library Transformations: Recovery of diverse plasmid or genomic libraries in E. coli before applying selection to maintain representation.

  • CRISPR Editing Workflows: Post-transformation outgrowth of edited E. coli to enable expression of selection markers prior to plating.

  • Phage Display/Phagemid Rescue: Enrichment and recovery of M13-based phagemids in E. coli prior to selection or amplification.

  • Blue-White Screening Preparation: Brief outgrowth of transformed E. coli to improve colony formation and reporter activity on indicator plates.

  • Antibiotic Selection Readiness: Allows time for antibiotic resistance gene expression in transformed E. coli before exposure to antibiotics.

  • Quality Control of Competent Cells: Recovery step for control plasmid transformations in E. coli to assess competency performance.

  • General Plasmid Cloning Workflows: Routine short-term culture of transformed E. coli prior to plating or mini-prep initiation.
