AAV•Tek AEX Buffer: AAV2 - Equilibration Buffer 24A is a ready-to-use buffer intended for equilibrating strong anion-exchange chromatography media prior to loading AAV2 samples. It is used to establish column conditions at defined pH and ionic strength appropriate for AAV2 processing and for preparing process solutions as specified in method instructions.

  • Column Equilibration (AEX): Prepares anion exchange columns for loading AAV2 by setting initial pH and conductivity conditions.

  • Column Conditioning/Stabilization: Establishes a stable baseline and consistent binding environment between runs.

  • Sample Preparation/Buffer Exchange: Dilutes or adjusts feed streams into AEX starting conditions prior to loading.

  • Impurity Wash Steps: Used in wash phases to remove host cell proteins, DNA, and process-related impurities while retaining AAV2 on the column.

  • Process Development: Supports method optimization and robustness studies for AEX-based AAV2 purification.

  • Scale-Up/Manufacturing: Enables consistent column equilibration across pilot and GMP production runs.

  • Re-Equilibration: Restores initial conditions between gradient steps or after column regeneration.

  • System Suitability/Validation: Used during system checks and validation runs to verify AEX performance for AAV2 workflows.

  • pH: 8.60 ± 0.10
  • Endotoxin: ≤0.5EU/mL
