Glycerol Solution is an aqueous polyol reagent used as a cryoprotectant and stabilizer in microbiology and molecular biology workflows. It is commonly employed in the preparation of frozen stocks of microbial cultures, including E. coli, to help mitigate ice crystal formation during low-temperature storage. The solution is miscible with water and compatible with a wide range of buffers and media.

Applications include preservation of bacterial and yeast isolates, dilution or stabilization of enzymes and proteins sensitive to freeze–thaw stress, and preparation of working stocks for routine laboratory use. Suitable for workflows that require consistent viscosity and hygroscopic properties without introducing additional salts or nutrients.

  • 50% (v/v) Glycerol
  • Cryopreservation of Microbial Stocks: Prepares long-term glycerol stocks of bacteria (e.g., Escherichia coli, Bacillus subtilis) and yeast by diluting 50% (v/v) Glycerol to 10–25% final.

  • Competent Cell Preparation and Electroporation: Uses 50% (v/v) Glycerol to make 10% glycerol washes/resuspensions for low-conductivity handling of E. coli and other bacteria.

  • Enzyme and Protein Stabilization: Adds 10–50% glycerol (from 50% (v/v) Glycerol stock) to storage buffers to prevent freezing and stabilize enzymes/proteins at −20 °C.

  • Gel Electrophoresis Sample Loading: Increases sample density by adding glycerol (from 50% (v/v) Glycerol) to DNA/RNA/protein loading buffers for reliable well loading.

  • Protein Purification and Storage Buffers: Incorporates glycerol (via 50% (v/v) Glycerol stock) to reduce aggregation and enhance protein stability during purification and storage.

  • PCR/qPCR Additive: Supplements reactions with small percentages of glycerol (prepared from 50% (v/v) Glycerol) to aid amplification of difficult templates and stabilize polymerases.

  • Cryoprotection in Structural Biology: Serves as a cryoprotectant (typically 10–30% from 50% (v/v) Glycerol) for protein crystals and samples prior to flash-freezing.

  • Microbial Carbon Source: Provides a defined carbon source in culture media (e.g., glycerol minimal media for Escherichia coli) by dosing from 50% (v/v) Glycerol to 0.2–2% final.

  • Phage and Viral Stock Stabilization: Protects bacteriophage and viral preparations during freezing by adding glycerol derived from 50% (v/v) Glycerol.
