LB Agar Plates with 2% Glucose and Carbenicillin-100 are solid microbiological media formulated for cultivation and selection of carbenicillin-resistant E. coli strains. The base is LB agar with added glucose (2%) as a readily metabolizable carbon source.

Carbenicillin is present at 100 µg/mL as the disodium salt to maintain selection for beta‑lactam–resistant, plasmid‑bearing transformants.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 1.5% Agar
  • 2.0% Cerelose Anhydrous Dextrose (Glucose)
  • 100µg/mL Carbenicillin Disodium Salt
  • Antibiotic Selection of AmpR Plasmids: Selects for carbenicillin-resistant transformants, enabling maintenance of AmpR plasmids in E. coli.

  • Post-Transformation Plating: Recovers and isolates colonies after heat-shock or electroporation of E. coli with carbenicillin selection.

  • Cloning with Lac-Based Promoters: 2% glucose suppresses basal expression from lac/tac/T7-lac promoters, aiding cloning of toxic or burdensome inserts in E. coli.

  • Clean Colony Isolation: Carbenicillin reduces ampicillin-type satellite colonies, improving colony picking accuracy from E. coli plates.

  • Plasmid Maintenance and Passaging: Sustains selective pressure during routine streaking and passaging of plasmid-bearing E. coli.

  • Colony Screening and PCR Prep: Supports reliable colony growth for downstream colony PCR and sequencing of E. coli clones.

  • CFU Enumeration Under Selection: Quantifies viable, plasmid-bearing E. coli by colony-forming unit counts on selective plates.

  • Library and Variant Recovery: Recovers combinatorial or mutagenesis libraries in E. coli while minimizing leaky expression during initial outgrowth.
