Manganese(II) chloride solution that provides a defined source of Mn2+ ions for laboratory use. Commonly used for trace metal supplementation in microbial and yeast media, enzyme cofactor studies, and preparation of minimal media and buffer systems where controlled manganese levels are required.

Applications include incorporation into trace element mixes for culture media, addition to biochemical assays evaluating manganese-dependent enzymes, and use in protocols that modulate divalent cation concentrations for growth and metabolism studies in bacteria such as E. coli and yeast.

  • 1M Manganese Chloride
  • PCR Optimization and Error-Prone PCR: Added as a Mn2+ cofactor to modulate DNA polymerase fidelity, enhance amplification of difficult templates, and increase mutation rates in error-prone PCR.

  • Reverse Transcription: Used to enable or improve RT performance on structured RNA templates by substituting or supplementing Mg2+ with Mn2+.

  • Nuclease and Restriction Enzyme Assays: Employed to activate or alter specificity of nucleases (e.g., DNase I) and certain restriction enzymes that accept Mn2+.

  • Glycosyltransferase Reactions: Provides essential Mn2+ cofactor for many glycosyltransferases in glycan synthesis and protein glycosylation assays.

  • Metalloenzyme Activity Assays: Supplies Mn2+ for manganese-dependent enzymes (e.g., MnSOD, arginase) to assess kinetics and cofactor requirements.

  • Bacterial Competent Cell Preparation: Included in transformation buffers to improve DNA uptake in Escherichia coli and Bacillus subtilis.

  • Microbial Culture Media Supplementation: Added as a trace metal source to support growth and metabolism in bacteria (e.g., E. coli, Bacillus spp.) and yeast fermentations.

  • Protein Crystallography and Structural Studies: Used to replace Mg2+ or stabilize metal-binding sites, aiding crystal formation and mechanistic trapping.

  • CRISPR and Nucleic Acid Editing Systems: Mn2+ supplementation can modulate activity and specificity of certain CRISPR-associated nucleases.
