Trypticase Soy Agar (TSA) plates are a general-purpose solid medium formulated with casein and soybean peptones, sodium chloride, and agar. This composition provides a balanced source of amino acids, peptides, and electrolytes suitable for the cultivation of a broad range of nonfastidious microorganisms under aerobic or facultative anaerobic conditions.

TSA plates are commonly used for routine cultivation, maintenance of stock cultures, environmental monitoring, bioburden assessment, and colony morphology observation of bacteria such as Escherichia coli, Staphylococcus aureus, Bacillus subtilis, Pseudomonas aeruginosa, and members of the Enterobacteriaceae.

  • 1.5% Tryptone
  • 0.5% Soytone
  • 0.5% Sodium Chloride
  • 1.5% Agar
  • General Microbial Cultivation: Broad-purpose growth medium for non-fastidious aerobic bacteria such as Escherichia coli, Staphylococcus aureus, and Pseudomonas aeruginosa.

  • Enumeration of Aerobic Plate Counts: Used for total viable counts in food, beverage, cosmetic, and environmental samples.

  • Environmental Monitoring: Employed as settle plates or contact plates (when appropriate) for air and surface monitoring in cleanrooms and laboratories.

  • Bioburden Testing: Quantifies microbial load on raw materials, in-process samples, and finished products in pharma and medical device QC.

  • Water and Utility Testing: Supports heterotrophic plate counts for potable water, purified water, and process utilities.

  • Clinical and Diagnostic Microbiology: Cultivation of non-fastidious pathogens and commensals including Bacillus subtilis, Enterococcus faecalis, and Salmonella enterica.

  • Isolation and Colony Purification: Streaking mixed cultures to obtain isolated colonies for downstream identification or testing.

  • Cultural Morphology and Maintenance: Observation of colony characteristics and routine maintenance of stock strains.

  • Sanitation and Disinfectant Efficacy Checks: Pre- and post-cleaning surface swab recovery to assess cleaning effectiveness.

  • Process and Personnel Monitoring: Finger dab and gown monitoring in aseptic processing environments.
