2XYT (Yeast extract Tryptone) Broth is a nutrient-rich liquid medium used for cultivation of Escherichia coli and other enteric bacteria in cloning, plasmid maintenance, and recombinant protein workflows. Its balanced composition provides amino acids, peptides, vitamins, and electrolytes suitable for high-density bacterial culture.

This medium can be used alone or supplemented with antibiotics and inducers as required by specific vectors or expression systems. It is suitable for routine propagation of plasmid-bearing E. coli strains and for preparing starter cultures, scale-up cultures, and downstream processing in molecular biology applications.

  • 1.6% Tryptone
  • 1.0% Yeast Extract
  • 0.5% Sodium Chloride

  • High-Density Culture of Escherichia coli: Rich medium for rapid growth and elevated cell density compared to LB.

  • Plasmid DNA Preparation: Cultivation for mini/midi/maxiprep workflows to increase plasmid yield.

  • Recombinant Protein Expression: Supports robust expression in T7 and other inducible systems in Escherichia coli.

  • Bacteriophage Propagation: Amplification of M13/phage display libraries and helper phage growth.

  • Transformation Outgrowth: Recovery of competent Escherichia coli post-transformation to express antibiotic resistance markers.

  • Cloning and Selection: General-purpose broth (and agar) for maintenance and selection of recombinant Escherichia coli strains.

  • Starter Cultures for Fermentation: Seed culture preparation for downstream bioprocessing.

  • Library Amplification: Growth of cDNA/genomic and plasmid libraries in Escherichia coli for screening and enrichment.
