YPD Agar Plates with 1.2 M Sorbitol are a nutrient-rich solid medium used for the cultivation and recovery of yeast, including osmotically sensitive cells and spheroplasts. The medium provides a balanced supply of amino acids, peptides, vitamins, and growth factors, with glucose as a readily metabolizable carbon source.

The addition of sorbitol establishes a high-osmolarity environment that supports cell wall–compromised yeast during transformation, plating, and recovery workflows. Agar serves as the solidifying agent for colony isolation and routine propagation.

  • 1% Yeast Extract
  • 2% Peptone
  • 2% Agar
  • 2% Glucose
  • 1.2M Sorbitol
  • Yeast Transformation Recovery: Supports post-transformation recovery of spheroplasted or electroporated yeast by providing osmotic stabilization for improved viability.

  • Spheroplast/Protoplast Plating: Enables plating and colony formation of cell wall–compromised yeast following lyticase/zymolyase treatment.

  • Cell Wall Mutant Maintenance: Maintains viability of yeast strains with cell wall defects or osmotic sensitivity under hyperosmotic conditions.

  • Osmotic Stress Assays: Facilitates assessment of growth, stress tolerance, and phenotype under high-osmolarity conditions.

  • Clonal Isolation and CFU Enumeration: Allows single-colony isolation and CFU counts of yeast requiring osmotic support.

  • Post-CRISPR/Genome Editing Recovery: Aids recovery and outgrowth of edited yeast cells that are temporarily cell wall–compromised.

  • Fungal Spheroplast Workflows: Suitable for plating spheroplasts of yeasts such as Saccharomyces and Candida species during genetic or cell biology studies.
