SGCAA Media with Galactose is a defined yeast medium for cultivation and maintenance of Saccharomyces cerevisiae. Galactose serves as the primary carbon source suitable for GAL-inducible workflows, yeast nitrogen base and casamino acids provide mineral nutrients and amino acids, and a phosphate buffer system helps maintain pH during growth. The medium is compatible with auxotrophic selection when supplemented with the appropriate dropout components.

  • 2.00% D-(+)-Galactose
  • 0.67% Yeast Nitrogen Base
  • 0.50% Casamino Acids
  • 0.54% Disodium Phosphate
  • 0.86% Monosodium Phosphate
  • Galactose-Inducible Expression in Yeast: Supports induction of GAL1/GAL10-driven constructs in Saccharomyces cerevisiae using 2.00% D-(+)-Galactose.

  • Yeast Cultivation and Maintenance: Provides a defined base (0.67% Yeast Nitrogen Base) with amino acids (0.50% Casamino Acids) for routine growth of laboratory yeast strains.

  • Protein Production in Yeast: Enables galactose-induced expression for recombinant protein and secreted protein workflows.

  • Metabolic and Regulatory Studies: Used to study galactose utilization pathways and relief of glucose repression in yeast.

  • Yeast Two-Hybrid and Reporter Assays: Facilitates galactose-dependent activation of GAL-promoter reporters in assay workflows.

  • Pre-Cultures and Carbon Source Shift Experiments: Employed for preculturing and switching from glucose to galactose to initiate induction.

  • Transformation Recovery and Plasmid Maintenance: Serves as a buffered recovery medium post-transformation prior to selection.

  • pH-Stable Yeast Growth: 0.54% Disodium Phosphate and 0.86% Monosodium Phosphate provide buffering for consistent culture pH.
