Tryptic Soy Agar Plates with 5% Sheep’s Blood are an enriched, non-selective medium used for the cultivation of a broad range of aerobic and facultative anaerobic bacteria and for observation of hemolytic reactions. The incorporation of defibrinated sheep’s blood provides the intact red cells and hemoglobin needed to visualize alpha, beta, or non-hemolytic phenotypes.

Typical applications include recovery and differentiation of organisms such as Streptococcus spp. and Staphylococcus aureus from clinical or environmental specimens, with incubation commonly performed at 35–37 °C in ambient air or 5% CO₂ as required by the organism.

  • 1.5% Tryptone
  • 0.5% Soytone
  • 0.5% Sodium Chloride
  • 5.0% Defibrinated Sheep's Blood
  • 1.5% Agar
  • Cultivation of Fastidious Organisms: Supports primary isolation and robust growth of fastidious bacteria from clinical and environmental specimens using a TSA base with 5% defibrinated sheep’s blood.

  • Hemolysis Differentiation: Enables visualization of alpha, beta, and gamma hemolysis patterns for presumptive identification of streptococci and other hemolytic species.

  • Throat and Respiratory Pathogen Isolation: Recovery and presumptive identification of beta-hemolytic streptococci such as Streptococcus pyogenes and alpha-hemolytic species like Streptococcus pneumoniae.

  • Staphylococcal Isolation: Isolation and assessment of hemolytic activity of Staphylococcus aureus from wound, blood, and respiratory specimens.

  • Listeria Screening: Observation of narrow-zone beta-hemolysis to aid in detection of Listeria monocytogenes from food and clinical samples.

  • Satellitism Test: Detection of growth enhancement (satellitism) of Haemophilus influenzae around Staphylococcus aureus streaks for presumptive identification.

  • Colony Morphology and Pigment Assessment: Evaluation of colony size, color, hemolysis, and odor for preliminary microbial identification.

  • Antimicrobial Susceptibility Screening: Qualitative disk diffusion screening for fastidious organisms when method and supplement guidelines permit.

  • Enumeration and Bioburden Testing: CFU counts and microbial load assessment in clinical, food, and environmental monitoring workflows.

  • Subculture and Maintenance: Routine subculture and short-term maintenance of clinical isolates requiring enriched, non-selective media.
