Mueller-Hinton II Cation-Adjusted Broth is a nutrient medium designed for standardized antimicrobial susceptibility testing by broth dilution and MIC determination with aerobic, nonfastidious bacteria, including Escherichia coli, Staphylococcus aureus, Pseudomonas aeruginosa, and related taxa. It provides amino acids, peptides, and carbohydrates from casein digest and beef extract, with soluble starch included to bind potentially inhibitory compounds.

The concentrations of divalent cations are adjusted with defined amounts of calcium and magnesium salts to establish consistent Ca2+ and Mg2+ levels appropriate for susceptibility testing protocols.

  • Antimicrobial Susceptibility Testing (AST): Standard medium for Kirby–Bauer disk diffusion; cation adjustment supports accurate results for aminoglycosides and tetracyclines against organisms like Escherichia coli, Staphylococcus aureus, and Pseudomonas aeruginosa.

  • Broth Microdilution MIC Testing: Used to determine minimum inhibitory concentrations of antibiotics for nonfastidious bacteria (e.g., E. coli, Enterococcus faecalis, P. aeruginosa).

  • Broth Macrodilution MIC/MBC: Supports tube dilution and bactericidal (MBC) assays for comparative potency and kill-curve assessments.

  • Quality Control of AST: Media and antibiotic performance verification with QC strains such as E. coli ATCC 25922, S. aureus ATCC 29213/25923, and P. aeruginosa ATCC 27853.

  • Growth of Nonfastidious Bacteria: General-purpose cultivation and maintenance in log phase for organisms like Staphylococcus aureus, Escherichia coli, and Enterococcus faecalis.

  • Colistin/Polymyxin MIC Testing: Cation-adjusted formulation supports reliable susceptibility testing for polymyxins against Gram-negative bacteria (e.g., E. coli, P. aeruginosa).

  • Time–Kill Kinetics Studies: Evaluates bactericidal activity of antimicrobials over time in standardized broth conditions.
