XLD Agar Plates are pre-poured plates containing XLD agar, a selective and differential medium used for the isolation and differentiation of enteric bacteria. It is used to recover and distinguish Salmonella and Shigella from clinical, food, and environmental samples based on carbohydrate fermentation and hydrogen sulfide production.

Typical colony reactions: Salmonella form red colonies with black centers; Shigella form red colonies without black centers; lactose/sucrose fermenters produce yellow colonies.

Recommended conditions: incubate at 35–37 °C for 18–24 h in ambient air. Storage: 2–8 °C, protected from light; allow plates to reach room temperature before use.

  • XLD Agar
  • Clinical Stool Cultures: Isolation and differentiation of Salmonella and Shigella from patient specimens.

  • H2S Production Detection: Presumptive identification of H2S-positive Salmonella via black-centered colonies.

  • Food and Dairy Testing: Screening food matrices for Salmonella contamination and differentiation from coliforms.

  • Environmental and Water Monitoring: Detection of enteric pathogens, especially Salmonella, in environmental samples.

  • Post-Enrichment Isolation: Subculture from Selenite F, Rappaport-Vassiliadis, or Tetrathionate broths to recover Salmonella/Shigella.

  • Fermentation Differentiation: Distinguishes xylose, lactose, and sucrose fermenters; helps separate coliforms like Escherichia coli from pathogens.

  • QC in Regulated Labs: Routine microbiological quality control for absence/presence of Salmonella in cGMP and industrial settings.

  • Proteus Swarm Suppression: Inhibits swarming of Proteus species to facilitate isolated colony recovery.
