SG-CAA Minus Leucine and Tryptophan is a galactose-based, defined yeast medium containing casamino acids with leucine and tryptophan omitted. Ammonium sulfate supplies inorganic nitrogen and phosphate salts provide buffering.

This formulation supports the cultivation of Saccharomyces cerevisiae and enables selection for LEU2 and TRP1 markers or prototrophy in strains or plasmids under galactose-utilizing or GAL promoter–driven conditions.

  • Yeast Two-Hybrid Screening: Selects for LEU2 and TRP1 markers (-Leucine, -Tryptophan) while supporting galactose-driven reporter activation.

  • Plasmid Maintenance and Selection: Maintains yeast plasmids carrying LEU2/TRP1 auxotrophic markers under dropout conditions.

  • GAL Promoter Induction: 2.0% D-Galactose enables induction of GAL1/10-driven gene expression in yeast.

  • Transformation Recovery and Selection: Supports growth and selection of transformed yeast on -Leu/-Trp media.

  • Replica Plating and Colony Screening: Facilitates dropout-based verification of auxotrophic markers and construct integrity.

  • Protein–Protein Interaction Assays: Enables interaction readouts in GAL-based yeast two-hybrid workflows.

  • CRISPR Editing Workflows: Provides defined selection for edited yeast strains using LEU2/TRP1 markers.

  • Growth and Fitness Assays: Defined nitrogen (0.5% ammonium sulfate) and phosphate buffer (0.54% disodium phosphate/0.86% monosodium phosphate) support reproducible phenotyping.

  • Library Construction and Screening: Supports selection and enrichment of yeast genomic or cDNA libraries under -Leu/-Trp conditions.

  • Diploid/Mating Selections: Enables selection of diploids and double auxotrophs using combined dropout markers.
