Brain Heart Infusion Agar Plates with Sucrose are a nutrient-rich solid medium for the cultivation and maintenance of a broad range of bacteria and yeasts. The formulation is based on Brain Heart Infusion and solidified with agar, with sucrose included as a fermentable carbohydrate source.

This medium is used in studies of carbohydrate utilization, acidogenicity, and extracellular polysaccharide formation by organisms such as Streptococcus mutans, Streptococcus sobrinus, and Enterococcus faecalis, as well as for general-purpose culture of fastidious and non-fastidious microbes.

  • 3.70% Brain Heart Infusion Broth
  • 1.50% Agar
  • 10.00% Sucrose
  • Isolation of Cariogenic Streptococci: Recovery and enumeration of Streptococcus mutans and Streptococcus sobrinus from saliva or dental plaque, leveraging sucrose-dependent mucoid/EPS-rich colony morphology.

  • Biofilm and EPS Screening: Assessment of sucrose-dependent biofilm and exopolysaccharide production by oral streptococci and staphylococci (e.g., Streptococcus mutans, Streptococcus sanguinis, Staphylococcus aureus).

  • Adherence Assays: Surface adherence/colonization tests on tooth analogs or discs for Streptococcus mutans, Streptococcus sanguinis, and Lactobacillus spp. under high-sucrose conditions.

  • Bacteriocin (Mutacin) Overlays: Indicator lawns and overlay assays for detection of mutacin/bacteriocin activity produced by Streptococcus mutans.

  • Antimicrobial Susceptibility on Mucoid Colonies: Spot and gradient assays against sucrose-induced biofilm-like growth of oral streptococci.

  • Maintenance of Fastidious Organisms: Routine cultivation of rich-medium–requiring bacteria (e.g., Streptococcus spp., Enterococcus faecalis, Listeria monocytogenes) on a non-selective, high-carbohydrate agar.

  • Phenotypic Differentiation: Observation of sucrose-driven mucoid/rough colony variants linked to EPS production in Streptococcus and Staphylococcus spp.

  • Genetic Manipulation Workflows: Recovery/selection plating of transformed or transconjugant oral streptococci requiring robust growth and sucrose-dependent phenotypes.
