CM Glucose Plates minus Lysine are defined agar plates formulated with glucose as the primary carbon source and ammonium sulfate as the inorganic nitrogen source, with no lysine added. The medium is intended for the maintenance and differentiation of strains based on lysine prototrophy or auxotrophy, including laboratory yeast such as Saccharomyces cerevisiae and bacteria such as Escherichia coli, when used in appropriate genetic contexts.

Applications include replica plating, strain verification, and selection workflows that rely on lysine omission. Plates are ready-to-use and suitable for routine microbiological procedures that require a defined glucose medium lacking lysine.

  • 2.0 % Cerelose Anhydrous Dextrose (Glucose)
  • 0.5% Ammonium Sulfate
  • Auxotrophic Selection (LYS2): Selects lys2Δ yeast transformants complemented with LYS2 on plasmids or integrations; only LYS+ colonies grow on lysine dropout with 2.0% glucose and 0.5% ammonium sulfate.

  • Plasmid/Integration Maintenance: Maintains selection pressure for LYS2-marked constructs in Saccharomyces cerevisiae on defined glucose plates lacking lysine.

  • Yeast One-/Two-Hybrid Assays (LYS2 Reporter): Selects activation of LYS2 reporters by growth on -Lys synthetic plates containing 2.0% glucose.

  • Replica Plating Screens: Identifies lysine auxotrophs vs prototrophs by differential growth on -Lys vs complete plates.

  • Tetrad Dissection and Segregant Genotyping: Scores meiotic segregation of LYS2/lys2 markers via growth on lysine dropout medium.

  • Gene Deletion/Marker Swap Workflows: Uses LYS2 as a selectable marker during construction and verification of engineered yeast strains.

  • Mutation/Reversion Rate Assays: Quantifies spontaneous reversion or suppression of lys2 mutations through growth on -Lys plates.
