500 mM EDTA, pH 8.0, Endotoxin Tested is a chelating solution that sequesters divalent metal ions such as Mg2+ and Ca2+. It is used in nucleic acid preservation and enzymatic reaction buffers to limit nuclease activity, in cell lysis and wash solutions, and in microbiology workflows with organisms such as E. coli where controlled chelation of cations is required. Endotoxin tested.

  • 500mM Ethylenediaminetetraacetic acid (EDTA)
  • Nucleic Acid Preservation (TE Buffer): Chelates Mg2+/Ca2+ to inhibit DNases/RNases, stabilizing DNA/RNA in Tris-EDTA storage buffers.

  • Plasmid DNA Preparation from Escherichia coli: Included in resuspension buffers to inactivate nucleases and weaken the outer membrane during alkaline lysis.

  • Quenching Enzymatic Reactions: Stops Mg2+-dependent enzymes (e.g., polymerases, ligases, restriction endonucleases, DNases) by chelating divalent cations.

  • Protein Extraction and Protease Control: Inhibits metalloproteases in cell lysis and purification buffers to protect proteins from degradation.

  • IMAC Resin Stripping: Removes Ni2+/Co2+ from immobilized metal affinity chromatography resins for regeneration.

  • Competent Cell and Transformation Workflows: Used to chelate surface-stabilizing cations, aiding permeability of Gram-negative cells like Escherichia coli.

  • Cell Culture Detachment (Trypsin-EDTA): Facilitates dissociation of adherent mammalian cells by chelating Ca2+/Mg2+ that mediate cell–substrate adhesion.

  • Bacterial Cell Lysis Support: Enhances lysozyme and detergent-mediated lysis of Gram-negative bacteria (e.g., Escherichia coli, Pseudomonas aeruginosa) by disrupting LPS stabilization.

  • Metal-Free Buffer Preparation: Removes trace divalent metals from reagents to control metal-dependent biochemical assays.

  • Tissue Decalcification: Demineralizes bone and calcified samples in histology while preserving biomolecular integrity.
