5-FOA Selective Plates are synthetic defined agar plates for growth of yeast and counter-selection against URA3 function. The medium is based on yeast nitrogen base without amino acids or ammonium sulfate, with glucose as the carbon source, ammonium sulfate as the inorganic nitrogen source, and a complete supplement mix to supply amino acids and bases; 5‑fluoroorotic acid enables selection of ura3 cells and loss of URA3-marked elements.

  • 0.17% Yeast Nitrogen Base w/o AA/AS
  • 0.13% AA CSM Mix
  • 2.00% Agar
  • 0.10% 5-fluorouracil-6-carboxylic acid monohydrate (5-FOA)
  • 0.50% Ammonium Sulfate
  • 2.00% Cerelose Anhydrous Dextrose (Glucose)
  • URA3 Counterselection in Yeast Genetics: Selects against URA3+ cells to isolate ura3 mutants or confirm loss of URA3 markers in Saccharomyces cerevisiae and related yeasts.

  • Marker Recycling/Pop-out Screening: Identifies successful excision of URA3 cassettes after homologous recombination or site-specific recombination steps.

  • Plasmid Shuffling: Counterselects URA3-marked plasmids to test essential gene alleles or swap functional variants.
