Description
Formulation

Storage

Glycerol solution supplied as a sterile, ready-to-use aqueous reagent for general laboratory applications in molecular biology, microbiology, and biochemistry. Suitable for use as a cryoprotectant, carbon source, viscosity modifier, and diluent in buffer and reagent preparation.

Common applications include preparation and storage of bacterial stocks (e.g., E. coli), stabilization of proteins and enzymes during handling, and procedures requiring a defined polyol content. The solution is clear and colorless, fully miscible with water, and packaged to maintain sterility.

For laboratory use only. Store under appropriate refrigerated conditions. Avoid contamination by using aseptic technique.

  • 10% (v/v) Glycerol
  • Preparation of Electrocompetent Cells: Ice-cold 10% (v/v) Glycerol washes to remove salts and maintain osmotic balance before electroporation of Escherichia coli and other bacteria.

  • Cryopreservation of Bacterial Stocks: Cryoprotectant for -80°C storage; mix cultures to 10–25% final glycerol for strains such as Escherichia coli and Staphylococcus aureus.

  • Protein and Enzyme Stabilization: Added at 10% (v/v) to storage and purification buffers to reduce aggregation and preserve activity during freeze–thaw cycles.

  • PCR/qPCR Additive: 5–10% glycerol can enhance amplification of GC-rich templates by stabilizing polymerases and reducing secondary structures.

  • Sample Loading for Electrophoresis: Provides density (typically ~10% glycerol in loading dyes) to ensure DNA/protein samples sink into wells for agarose or SDS-PAGE.

  • Molecular Cloning Workflows: Included at ~10% in enzyme storage or reaction buffers to stabilize ligases and restriction enzymes.

  • Density Gradient Preparation: Base solution for preparing glycerol gradients (e.g., 10–40%) for ribosome, protein, or nucleic acid fractionation.

  • Membrane Protein Handling: 5–10% glycerol in purification buffers to stabilize membrane proteins during extraction and chromatography.
