LB Broth is a nutrient medium used for the cultivation and maintenance of laboratory strains of E. coli and other enteric bacteria. It provides peptides, amino acids, vitamins, and electrolytes suitable for routine propagation in shake flasks and culture tubes.

LB Broth is commonly used for starter and overnight cultures, preparation of competent cells, and propagation of recombinant strains when supplemented with appropriate antibiotics. Sterilize by autoclaving and use under aseptic conditions.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • Routine Cultivation of Escherichia coli: General growth of Escherichia coli and other non-fastidious bacteria for laboratory workflows.

  • Plasmid Cloning and Propagation: Supports growth of recombinant strains for amplification of plasmid DNA.

  • Transformation Recovery: Provides a rich medium for post-heat shock or electroporation recovery of E. coli transformants.

  • Antibiotic Selection in Liquid Culture: Used with antibiotics to select for plasmid-bearing bacterial populations in broth.

  • Starter Cultures for Protein Expression: Generates robust inocula for expression studies in E. coli.

  • Overnight Cultures for DNA Prep: Grows cells for plasmid miniprep/midiprep workflows.

  • Preparation of Glycerol Stocks: Cultivates cells prior to cryopreservation for long-term strain storage.

  • Growth Curves and OD Calibration: Used for measuring bacterial growth kinetics and spectrophotometer calibration.

  • Strain Maintenance and Passaging: Routine subculturing to maintain laboratory E. coli strains.
