LB Agar is a solid nutrient medium for routine cultivation and plating of nonfastidious bacteria, including E. coli. It supplies peptides, amino acids, vitamins, and sodium chloride for cell growth and osmotic balance, and is solidified with agar to enable colony formation.

Applications include streaking, colony isolation, and maintenance of laboratory strains. The medium can be supplemented with antibiotics, inducers, or indicators as required for selection or screening; no antibiotics or carbohydrates are included unless added.

Typical preparation: dissolve the dehydrated medium in purified water, adjust to pH 7.0 ± 0.2 at 25 °C, autoclave at 121 °C for 15 min, then cool to 50–55 °C before pouring plates.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 1.5% Agar
  • Bacterial Cultivation and Maintenance: General-purpose growth medium for nonfastidious bacteria such as Escherichia coli, Bacillus subtilis, and Pseudomonas aeruginosa.

  • Colony Isolation: Streaking for single colonies to obtain clonal populations and assess colony morphology.

  • CFU Enumeration: Plating serial dilutions to quantify viable cells in cultures or samples.

  • Antibiotic Selection: Selection and maintenance of plasmid-bearing transformants on LB agar supplemented with antibiotics.

  • Cloning and Transformation Workflows: Recovery and plating of transformed Escherichia coli; compatible with blue-white screening (with X-gal/IPTG).

  • Colony Picking for Downstream Analysis: Isolating colonies for colony PCR, Sanger sequencing, and plasmid minipreps.

  • Phage Workflows: Base agar for bacteriophage plaque assays with soft agar overlays.

  • Teaching and QC Applications: Routine microbiology instruction, sterility checks, and general laboratory quality control.
