LB (Luria-Bertani) Broth is a nutrient-rich liquid medium used for routine cultivation of nonfastidious bacteria, including E. coli. Tryptone and yeast extract supply peptides, amino acids, nucleotides, vitamins, and minerals, while sodium chloride maintains osmotic balance.

Common applications include propagation of recombinant strains, maintenance of plasmids without selection, and preparation of starter cultures for downstream workflows involving E. coli.

  • 1.0% Tryptone
  • 0.5% Yeast extract
  • 1.0% Sodium Chloride

  • Bacterial Cultivation: Routine growth and maintenance of Escherichia coli and other non-fastidious bacteria.

  • Plasmid Propagation: Amplification of plasmid DNA for miniprep/midiprep and cloning workflows.

  • Recombinant Protein Expression: Culture medium for expression of recombinant proteins in Escherichia coli.

  • Antibiotic Selection: Selection and maintenance of transformants using antibiotics (e.g., ampicillin, kanamycin).

  • Transformation Recovery: Outgrowth after chemical or electroporation transformation of Escherichia coli.

  • Starter/Overnight Cultures: Seed cultures for downstream fermentation, expression, and assay setups.

  • Glycerol Stock Preparation: Growth prior to cryopreservation of bacterial strains.

  • Growth Curves (OD600): Monitoring bacterial growth kinetics and media optimization studies.

  • Bacteriophage Work: Propagation and titration of phages that infect Escherichia coli.

  • Colony Inoculation: Picking colonies into liquid culture for screening and verification assays.
