M9 Minimal Salts Agar Plates are a defined minimal medium formulated for cultivation and characterization of glucose-utilizing bacteria under controlled nutrient conditions. They are commonly used for genetic and physiological studies with E. coli and for experiments that assess growth requirements, auxotrophies, and carbon source utilization on a solid surface.

Glucose provides the primary carbon and energy source, thiamine supplies vitamin B1 required by certain laboratory strains, and magnesium and calcium furnish essential divalent cations. The agar base enables colony isolation, replica plating, and comparative growth assays, and the medium can be supplemented with amino acids, antibiotics, or alternative carbon sources as needed for specific selection or phenotype testing.

  • 1.0% Cerelose Anhydrous Dextrose (Glucose)
  • 1mM Thiamine
  • 0.2mM Magnesium Sulfate
  • 0.1mM Calcium Chloride
  • 1.5% Agar
  • Auxotroph/Prototroph Selection: Defined medium (1X M9 Salts, 1.0% glucose, 1 mM thiamine, 0.2 mM MgSO4, 0.1 mM CaCl2, 1.5% agar) for screening nutritional mutants in E. coli.

  • Defined-Condition Growth Studies: Controlled colony growth and physiology of enteric bacteria (e.g., E. coli, Salmonella enterica) under minimal nutrients.

  • Carbon Source Utilization Assays: Assess glucose-based metabolism, catabolite repression, and pathway analysis in E. coli on minimal agar.

  • Genetic Selection with Metabolic Markers: Selection of recombinants using markers (e.g., his, leu, lac) by adding or omitting specific supplements.

  • Antibiotic Selection of Transformants: Plating of plasmid-bearing E. coli on minimal medium supplemented with appropriate antibiotics/nutrients.

  • Bacteriophage Assays: Plaque assays with E. coli hosts under defined nutrient conditions for phage-host interaction studies.
