Phosphate-buffered saline containing a nonionic surfactant and a carrier protein is an isotonic wash and dilution buffer for immunoassays and general laboratory workflows. The buffer maintains physiological pH and osmolarity during sample handling and wash steps.

The surfactant promotes efficient wetting and helps disrupt weak hydrophobic interactions during washes, while the protein component provides a background matrix that reduces nonspecific adsorption of antibodies and other proteins to plastics and membranes. Applications include ELISA plate washing, Western blot wash and block steps, immunofluorescence washes, and preparation of antibody diluents.

  • 1X PBS Buffer with 0.05% Tween-20 (v/v) and 0.1% Bovine Serum Albumin (BSA)
  • Western Blotting: Blocking and wash buffer to reduce non-specific antibody binding on membranes.

  • ELISA: Plate blocking, sample/antibody diluent, and wash steps to minimize background signal.

  • Immunohistochemistry/Immunocytochemistry: Antibody diluent and wash buffer to improve specificity in tissue and cell staining.

  • Immunofluorescence Microscopy: Reduces non-specific binding and background during antibody staining.

  • Flow Cytometry: Staining wash and antibody diluent to decrease cell-surface non-specific interactions.

  • Dot Blot/Colony Blot: Membrane blocking and washing to enhance signal-to-noise.

  • Bead-Based Immunoassays: Magnetic/agarose bead blocking and washes for cleaner binding assays.

  • Surface Blocking: Pre-blocking microplates, tubes, and biosensor surfaces to prevent non-specific adsorption.

  • Antibody/Protein Dilution: Stabilizing diluent for primary/secondary antibodies and proteins.

  • Microbial Antigen Assays: Used in ELISAs and blots involving Escherichia coli or Staphylococcus aureus antigens to reduce background.
