20X PBS with 1% Tween-20 is a concentrated phosphate-buffered saline containing a nonionic surfactant. When diluted 1:20 with water, it yields 1X PBS-T with 0.05% Tween-20 suitable for routine wash steps and buffer preparation in applications such as western blotting, ELISA, and immunostaining.

This formulation provides phosphate buffering and physiological saline upon dilution, with Tween-20 aiding in gentle surface-active rinsing of membranes, plates, and slides. For laboratory use only.

  • 2.7M Sodium Chloride
  • 54.0mM Potassium Chloride
  • 86.0mM Sodium Phosphate Dibasic
  • 28.0mM Potassium Phosphate Monobasic
  • 1.0% Tween-20
  • Western Blotting: Used as a membrane wash buffer to reduce nonspecific protein–antibody interactions during transfer and probing.

  • ELISA: Serves as a plate wash buffer to minimize background and improve signal-to-noise in antibody-based detection.

  • Immunohistochemistry/Immunocytochemistry: Employed for slide and cell washing (often after fixation/permeabilization) to reduce nonspecific binding in staining workflows.

  • Immunofluorescence Microscopy: Used for washing coverslips and samples to preserve antigen–antibody complexes while limiting background fluorescence.

  • Flow Cytometry (Fixed/Permeabilized Cells): Applied as a wash buffer during intracellular staining to decrease nonspecific antibody binding and cell clumping.

  • Antibody Dilution/Incubation Buffer: Provides a surfactant-containing diluent that stabilizes antibodies and reduces hydrophobic adsorption when combined with protein blockers.

  • Bead- and Plate-Based Immunoassays: Utilized for washing magnetic/latex beads and microplates (e.g., multiplex assays) to limit nonspecific interactions.

  • Immunoprecipitation/Co-IP Washes: Incorporated in wash steps to lower nonspecific protein binding while maintaining isotonic, buffered conditions.
