This salt–polyethylene glycol solution is used to precipitate and concentrate bacteriophages from clarified culture supernatants. It enables polyethylene glycol–mediated virion precipitation and recovery by centrifugation.

Common applications include processing filamentous coliphages such as M13 and fd, as well as some tailed phages, prior to plaque assays, genomic DNA extraction, or density‑gradient purification. It is routinely employed in workflows involving phages propagated in E. coli.

  • Bacteriophage Precipitation and Concentration: PEG/NaCl-mediated precipitation of M13, f1, T7, λ, and P1 phage from Escherichia coli cultures and lysates for downstream amplification or storage.

  • Phage Display Workflows: Concentration and cleanup of filamentous phage after helper phage rescue and prior to panning or titering.

  • Viral Particle Concentration from Cell Culture: Precipitation of enveloped and non-enveloped viruses from supernatants to increase titer for transduction or assay.

  • Environmental Virome Enrichment: Concentration of bacteriophages from water, wastewater, or soil extracts for metagenomics and plaque assays using hosts such as Escherichia coli and Pseudomonas spp.

  • Phage-Mediated Transduction Workflows: Preparation and concentration of transducing lysates (e.g., P1) for genetic exchange in Escherichia coli.

  • Macromolecule Precipitation: PEG-assisted precipitation of nucleic acids and protein complexes from dilute solutions for downstream purification.

  • Phage DNA Preparation: Concentration of phage particles prior to ssDNA extraction for cloning or sequencing.
