60% (w/v) Sucrose Solution is a high-density, osmotically active aqueous solution used for density gradient and cushion centrifugation, and for osmotic stabilization of protoplasts and spheroplasts from yeast and bacteria such as E. coli.

Typical applications include preparation of step or continuous sucrose gradients for fractionation of organelles, membranes, ribonucleoprotein complexes, and viruses; cushioning for pelleting sensitive particles; and adjusting sample buoyant density in molecular biology, microbiology, and cell biology workflows.

Use as supplied. Store at 2–8 °C. Bring to room temperature and mix gently before use. Avoid dilution with hypotonic solutions that may reduce osmotic support.

  • 60% (w/v) Sucrose Solution
  • Density Gradient Centrifugation: Used with 60% (w/v) Sucrose Solution to form step or continuous gradients for separation of organelles, membranes, viruses, and macromolecular complexes.

  • Sucrose Cushion Ultracentrifugation: Serves as a high-density cushion to concentrate and purify viral particles, ribosomes, and vesicles during high-speed spins.

  • Virus and Vector Purification: Utilized to purify and concentrate viral vectors and bacteriophages by layering samples over sucrose cushions or gradients.

  • Polysome and Ribosome Profiling: Provides defined density layers for resolving monosomes, polysomes, and ribosomal subunits in translational studies.

  • Organelle Isolation: Used to fractionate mitochondria, lysosomes, endosomes, and plasma membranes based on buoyant density.

  • Sample Density Adjustment: Serves to increase sample density for clean layering onto gradients, minimizing mixing and improving band resolution.

  • Osmotic Stabilization of Protoplasts/Spheroplasts: Provides osmotic support during cell wall removal and gentle lysis of Escherichia coli, Bacillus subtilis, and Staphylococcus aureus.

  • Electron Microscopy and Cryoprotection: Utilized as a dense, osmoprotective medium for tissue handling, fixation workflows, and density matching prior to imaging.

  • Membrane Protein Preparation: Provides a gentle, non-ionic environment to stabilize membranes and membrane protein complexes during isolation.

  • Exosome and EV Isolation: Used in sucrose cushions/gradients to enrich extracellular vesicles with high purity and integrity.
