SM Buffer is a Tris-based salt buffer used for bacteriophage dilution, storage, and elution, and for preparing lysates or suspensions for plaque assays. Its composition provides buffering capacity, ionic strength, divalent cations, and a protein stabilizer to support particle stability and reduce surface adsorption during routine handling.

Suitable for workflows involving E. coli phages and other enteric bacteriophages, including preparation of high-titer lysates, elution from agar plugs or membranes, and serial dilutions for plaque enumeration.

  • 100mM Sodium Chloride
  • 10mM Magnesium Sulfate
  • 0.01% Gelatin
  • 50mM Tris-HCl
  • Bacteriophage Storage and Stabilization: Preserves phage infectivity using 50 mM Tris-HCl, 100 mM NaCl, 10 mM MgSO4, and 0.01% gelatin to maintain pH, ionic strength, and reduce surface adsorption.

  • Phage Dilution and Titering: Used for serial dilutions prior to plaque assays, ensuring consistent ionic conditions for accurate titers.

  • Plaque Assays: Employed to resuspend plaques and prepare phage stocks for enumeration and isolation.

  • Elution from Agar or Filters: Facilitates elution of phage from agar plugs, soft agar overlays, and membrane filters.

  • Phage Lysate and Pellet Resuspension: Resuspends PEG/NaCl-precipitated or centrifuged phage and is compatible with downstream CsCl gradients.

  • Phage DNA Extraction Prep: Serves as a stabilizing buffer for phage particles before nuclease treatments and genome isolation.

  • Phage Display and Library Handling: Maintains phage library stability during amplification, storage, and panning workflows.

  • Transport Medium for Phage Samples: Stabilizes environmental and clinical phage samples during collection and shipment.

  • Transduction Workflows: Storage and handling of P1, P22, and related lysates used for generalized transduction in Escherichia coli, Salmonella enterica, and Staphylococcus aureus.

  • Host Range and Infection Assays: Diluent for setting MOI and preparing inocula with phages infecting Escherichia coli and Pseudomonas aeruginosa.
