SOC Medium is a nutrient-rich, non-selective broth formulated for the short-term recovery and outgrowth of transformed E. coli following heat-shock or electroporation. It contains enzymatic digest of casein and yeast extract as sources of peptides, amino acids, vitamins, and growth factors, along with sodium and potassium salts to maintain osmotic balance.

Magnesium salts supply divalent cations commonly used during transformation workflows, and dextrose provides a readily metabolizable carbon source for rapid post-transformation outgrowth. SOC is suitable for use prior to plating on selective media when working with plasmid-bearing E. coli strains.

  • 2.0% Tryptone
  • 0.5% Yeast Extract
  • 10mM Sodium Chloride
  • 2.5mM Potassium Chloride
  • 10mM Magnesium Chloride
  • 10mM Magnesium Sulfate
  • 20mM Cerelose Anhydrous Dextrose (Glucose)
  • Post-Transformation Recovery (Heat Shock): Short-term outgrowth of chemically competent Escherichia coli after heat-shock to improve viability and plasmid uptake.

  • Post-Transformation Recovery (Electroporation): Gentle recovery of electrocompetent Escherichia coli following electroporation to maximize survival and transformant yield.

  • Maximizing Transformation Efficiency: Enhances plasmid transformation efficiency in E. coli during the first hour of outgrowth prior to selection.

  • Antibiotic Resistance Expression: Allows expression of antibiotic resistance markers in transformed E. coli before plating on selective media.

  • Competent Cell QC: Used in control reactions to assess and benchmark the efficiency of competent E. coli cells.

  • Library Cloning Workflows: Recovery and expansion of diverse plasmid or gDNA libraries in E. coli immediately post-transformation.
