YPD Broth with 1 M Sorbitol is a rich, nonselective medium for the cultivation of yeasts, including Saccharomyces cerevisiae and other laboratory and industrial strains. The sorbitol component provides defined osmotic support suitable for handling osmotically sensitive cells during procedures such as transformation, spheroplast preparation, and cell wall stress studies.

The medium supplies a readily metabolizable carbon source along with complex nitrogen, vitamins, and growth factors from peptone and yeast extract. The 1 M sorbitol concentration maintains osmotic balance to help preserve cell integrity in workflows that involve transient cell wall perturbation or high-osmolarity conditions.

  • Yeast Cultivation: Rich medium (2.0% Peptone, 1.0% Yeast Extract, 2.0% Glucose) for routine growth and propagation of yeast strains.

  • Post-Transformation Recovery: 1M Sorbitol provides osmotic stabilization for recovery after lithium acetate/PEG or electroporation of yeast.

  • Spheroplasting and Regeneration: Supports cell wall digestion and subsequent regeneration with osmotic support.
