YPD (Yeast extract Peptone Dextrose) Agar is a nutrient-rich solid medium for the cultivation and maintenance of yeasts, including Saccharomyces cerevisiae. Glucose serves as the primary carbon source, yeast extract provides vitamins, minerals, and growth factors, and peptone supplies peptides and amino acids; agar provides a firm surface for colony isolation and routine culture work.

Applications include nonselective growth, strain propagation, replica plating, and general phenotypic assays for yeast and other fungi in microbiology and molecular biology workflows.

  • 1.0% Yeast extract
  • 2.0% Peptone
  • 1.65% Agar
  • 2.0% Cerelose Anhydrous Dextrose (Glucose)
  • Yeast Cultivation and Propagation: Rich, non-selective medium for robust growth of laboratory yeasts on solid plates.

  • Colony Isolation and Enumeration (CFU): Supports single-colony isolation, counting, and purification of yeast strains.

  • Strain Maintenance: Routine streaking and passaging to maintain viable yeast stocks on solid media.

  • Transformation Recovery: Non-selective recovery medium after chemical or electroporation-based yeast transformations.

  • Replica Plating and Patching: Transfer of colonies for genetic screens, phenotype comparisons, and library handling.

  • Mating Assays: Crossing of yeast strains and subsequent growth to assess mating efficiency and zygote formation.

  • Phenotypic Screening: Assessment of growth under temperature, stress, or drug conditions on a rich background.

  • Starter Plate Preparation: Generation of robust inocula for subsequent liquid culture in rich media.
